New analytical tool reveals massive DNA damage caused by CRISPR/Cas9 gene editing

New research from Chinese scientists shows that CRISPR/Cas9 gene editing causes massive damage to the genome, much of which would have been missed by the analytical tools used so far.

In a previous study, the same authors described a novel DNA sequencing procedure, which they called a “primer-extension-mediated sequencing assay” or PEM-seq. By applying this procedure, they found that DNA double-strand breaks (DSBs) brought about by the CRISPR/Cas9 gene-editing tool could lead to unintended chromosomal translocations and large deletions.

In their latest follow-up report, the researchers describe a new computer program, which enabled them to analyse the PEM-seq data to greater depth than previous programs had allowed. They used this program to analyse real sequence data from their own new experiments, as well as previous ones, following CRISPR/Cas9 gene editing in mouse and human cells.

In gene editing, while the initial double-strand break made by the DNA “scissors” or gene-editing tool can be targeted to a given location, the subsequent DNA repair that makes the “edit” is performed by the cell’s own repair mechanisms and is not controllable or precise. The researchers analysed the outcomes of the gene edit – and found what they call “tremendous deleterious DSB repair byproducts of CRISPR/Cas9 editing”.

The unintended outcomes or genetic errors ranged from unintended small insertions or deletions (indels) to large deletions, plasmid (gene-editing tool delivery vehicle) integrations, and chromosomal translocations.

The researchers wrote, “Our findings provide an extra dimension for genome editing safety besides off-targets” – the well documented unintentional DNA damage at locations of the genome that were not targeted for editing. They added, “Caution should be exercised to avoid not only off-target damages but also deleterious DSB repair byproducts during genome editing.”

Worryingly, these unintended deleterious outcomes of gene editing were not overcome even when a high-fidelity CRISPR/Cas tool was used. Furthermore, when components of the DNA repair machinery were inhibited in an attempt to force a specific gene sequence modification outcome (known as SDN-2), this resulted in large deletions, large insertions, and DNA cuts around the start site of genes.

Collectively, these findings graphically highlight just how little control the gene editor has over the DNA repair processes and its outcomes, following the formation of the DSB by the CRISPR/Cas gene-editing tool.

Relevance to plant gene editing

How are the findings of the study relevant to plant gene editing? London-based molecular geneticist Dr Michael Antoniou explained, “The mechanisms of gene editing and the subsequent DNA repair processes are the same in animal and plant cells. Given that fact, the implications of the new research are clear. The genome sequencing analysis conducted by the scientists shows that all analyses of gene-edited plants done thus far need to be re-evaluated with this new tool to get a more complete picture of the DNA damage arising from gene editing.”…


Richard Ebright: The WHO Coronavirus Investigation Members Were “participants in disinformation”

Molecular biologist Dr. Richard H. Ebright, PhD is one of the twenty six world scientists who signed the Open Letter: “Call for a Full and Unrestricted International Forensic Investigation into the Origins of COVID-19”. A document released last March 4 by the Wall Street Journal and Le Monde that reignited the debate over the pandemic’s origin after the WHO-convened mission to Wuhan.

Board of Governors Professor of Chemistry and Chemical Biology at Rutgers University, Dr. Richard H. Ebright, PhD, is also Laboratory Director at the Waksman Institute of Microbiology and serves as project leader on two National Institutes of Health research grants.

Dr. Richard Ebright received his AB in Biology and his PhD in Microbiology and Molecular Genetics from Harvard University. He has more than one hundred sixty publications and more than forty issued and pending patents. He is member of the American Academy of Arts and Sciences, and a Fellow of the American Association for Advancement of Science, the American Academy of Microbiology, and the Infectious Diseases Society of America.

Dr. Ebright is a member of the Institutional Biosafety Committee of Rutgers University and the Antimicrobial Resistance Committee of the Infectious Diseases Society of America. He is also been a member of the Working Group on Pathogen Security of the state of New Jersey, and the Controlling Dangerous Pathogens Project of the Center for International Security Studies. He was a founding member of the Cambridge Working Group, which advocated for biosafety, biosecurity, and risk-benefit reviews for gain-of-function research on potential pandemic pathogens.

Dr. Ebright, you are one of the 26 scientific signers of the Open Letter that stopped the release of the WHO-convened mission Interim Report and has reopened the CoV-2 origin debate. Do you think the final report of this WHO/China joint team is going to shut down COVID origin controversy?


The Open Letter explains in detail the structural and functional limitations of the WHO-China Wuhan collaborative team: exclusive Chinese field work, lack of complete access to lab installations or databases, consensus process in report making…

A credible investigation would have had Terms of Reference that: 1) Acknowledged the possibility of laboratory origin, 2) Ensured access of investigators to records, samples, personnel, and facilities at the Wuhan laboratories that handle bat SARS-related coronaviruses, 3) Enabled collection of evidence, not mere meet-and-greet photo-ops, 4) Authorized an investigation of months, not mere days. And 5) A credible investigation also would have had conflict-of-interest-free investigators, not persons who were subjects of the research and/or closely associated with subjects of the investigation.

You have said several times that this WHO mission was literally “a charade”.

Yes, its members were willing –and, in at least one case, enthusiastic– participants in disinformation.

The pre-negotiated “Terms of Reference” for the WHO study did not even acknowledge the possibility of a laboratory origin of the virus and did not even mention the Wuhan Institute of Virology (WIV), the Wuhan Center for Disease Control (CDC) or the Wuhan Institute of Biological Products.

Regarding the inspection personnel, at least one member of the WHO mission team, Ecohealth Alliance President Dr. Peter Daszak, seems to have conflicts of interest that should have disqualified him from being part of an investigation of the origins of the COVID-19 pandemic.

Yes. Daszak was the contractor who funded the laboratory at WIV that potentially was the source of the virus (with subcontracts from $200 million from the US Department of State and $7 million from the US National Institutes of Health), and he was a collaborator and co-author on research projects at the laboratory.

The Open Letter also lists the elements of “What a full investigation should look like”. So again, Dr. Ebright, if you where in charge of a forensic team to investigate in Wuhan the origins of the pandemic: what would you like to see first? What kind of questions would you ask and to who?

A credible forensic investigation would require access to records, samples, personnel, and facilities at WIV, the Wuhan CDC and the Wuhan Institute of Biological Products. It would entail inspection of electronic and paper records, inspection of freezer and refrigerator samples, environmental sampling of facilities, and confidential interviews with personnel–including previous and current construction, maintenance, janitorial, disposal, security, animal-facility, laboratory, and administrative personnel.

Whenever asked if this virus could have leaked from the Wuhan Institute of Virology, your answer has been: “Yes (…) This cannot and should not be dismissed”. A study on COVID origin of Dr. Steven Quay, another signer of the Open Letter, calculates the lab hypotheses to be a 99.8% probability.

At this point in time, there is no secure basis to assign relative probabilities to the natural-accident hypothesis and the laboratory-accident hypothesis.

But given the fact that lab leak is a possibility, what chance do you give for a natural zoonotic origin of this pandemic? 

At this point in time, all scientific data related to the genome sequence of SARS-CoV-2 and the epidemiology of COVID-19 are equally consistent with a natural-accident origin or a laboratory-accident origin…..

Blackmail: Hunter Biden Says Laptop May Have Been His . . . Or The Russians Did It

In a  bizarre interview, President Joe Biden’s son Hunter Biden finally acknowledged that the infamous laptop may indeed be his . . . or the creation of Russian intelligence.  After offering detailed accounts going back years on his drug addiction and abuse, Biden seems incapable of remembering whether he had this laptop with thousands of files, including personal and embarrassing images of his having sex and doing drugs.

In his interview with CBS’s Tracy Smith, Biden is finally asked about the lap — which is an improvement over other media including a recent interview by NPR.

In the interview, Biden was asked about the laptop. He answered cryptically: “There could be a laptop out there that was stolen from me. It could be that I was hacked. It could be that it was the — that it was Russian intelligence.”

There was not a serious pushback on that incomprehensible answer like “you can remember details from your period of addiction going back 20 years but you cannot remember this laptop?”  Or how about this: “even if you cannot remember your own laptop, you have seen the pictures and emails. Are those authentic pictures and emails?”

Hunter Biden remains in a protected media space on questions about the specific emails and photos, including emails showing raw influence peddling worth millions.

Instead, crickets.


National Public Radio issued a correction after running a false statement about the laptop of Hunter Biden in a story about Biden’s recent memoir, “Beautiful Things.”  The article by NPR senior editor and correspondent Ron Elving stated categorically that the laptop story was discredited by news organizations. It was later compelled to correct that false statement but still has language casting doubt on the story and evades glaring contradictions in Biden’s book and his interview.  Moreover, NPR continues to run false claims from prior controversies.

The NPR story originally stated that “The laptop story was discredited by U.S. intelligence and independent investigations by news organizations.” That is entirely and demonstrably false. Anyone who has done a modicum of research would know that neither Hunter nor President Joe Biden have ever denied that this was Hunter Biden’s laptop or that the files and pictures were genuine. They have refused to specifically respond to the contents, a position enabled by the media which has maintained a protective cocoon around the Bidens. This included the blocking of the story by Big Tech companies and a virtual blackout on the story before the election….


Thus the media ensures that the blackmail control of the biden family continues.

Rep. Marjorie Taylor Greene Introduces Proposal to Bar ‘Vaccine Passports’

Rep. Marjorie Taylor Greene (R-Ga.) introduced a proposal on April 1 that would ban COVID-19 vaccination mandates as well as so-called vaccine passports.

The We Will Not Comply Act would ban documents that show who has been vaccinated or recently tested negative for the CCP virus. The concept of requiring certification has been rejected by pro-privacy groups such as the American Civil Liberties Union (ACLU).

Greene’s measure likely won’t be taken up by the Democratic-controlled House of Representatives, and if it is, it will be unlikely to pass. It would “prevent any business engaged in interstate commerce from discriminating against a person based on their COVID-19 vaccine status,” cut off federal funding for a vaccine mandate on employees, a vaccine mandate for students attending schools, and mandates on organizations or sport—or “any person who expresses religious objection,” according to a statement from her office.

It would also prohibit the government from requiring a CCP virus vaccine to get a U.S. passport and “prevent Airline companies from denying someone from flying based on their COVID-19 vaccination status,” as well as “create the ability to sue if a person has been discriminated against on the basis of their vaccination status or mask compliance,” her office wrote.

The idea of such a passport has been publicly floated by large corporations while anonymous reports this week suggested that the Biden administration was working on such a proposal. White House press secretary Jen Psaki, in responding to those reports, said the administration will instead “provide guidance” on the effort to develop vaccine passports or similar systems.

“There is currently an interagency process that is looking at many of the questions around vaccine verification, and that issue will touch many agencies, as verification is an issue that will potentially touch many areas of society,” she said.

“That’s guidance we’ll provide. A determination or development of vaccine passport or whatever you want to call it will be driven by the private sector. Ours will more be focused on guidelines that can be used as a basis and there are a couple of key principles that we are working from.”

Meanwhile, New York state has proposed its own “Excelsior Pass,” which would be used in large-scale venues such as Madison Square Garden. The plan has been criticized by civil liberties groups. Israel set one up in February to grant people access to gyms and hotels, while Iceland now uses a passport to allow foreign travel, and Saudi Arabia has an app-based passport for people who are inoculated.

Greene also introduced legislation that would eliminate the salary of National Institute of Allergy and Infectious Diseases (NIAID) chief Dr. Anthony Fauci, who her office said is being paid more than $430,000 per year.

The measure also has virtually no chance of advancing, but it seeks “to conduct a study about the correspondence, financials, and policy memos inside the NIAID before COVID through the end of this year. This will allow us to see what Fauci and the NIAID knew, when they knew it, what they spent money on, and how the agency responded to the virus.”


Fauci’s Orders: Children Not Allowed To Play Together Unmasked Until They’re All Vaxxed

While appearing on a Sunday show, Anthony Fauci made yet another controversial COVID-19 health dictate that seems to go against the science: Children are not allowed to play together while unmasked unless they have all been vaccinated.

When asked about children playing together in groups, Fauci said “The children can clearly wind up getting infected,” and, therefore, When the children go out into the community, you want them to continue to wear masks when they’re interacting with groups or multiple households.”

Fauci did, however, offer a ray of hope for America’s children: They may be able to hit the jungle gym unmasked as soon as this summer. Apparently not wanting to allow too much hope, Fauci simply acknowledged, it is “conceivable that will be possible.” Should the current vaccination rate continue, he explained, then there is a “good degree of flexibility during the summer … with things like camps.”

Fauci did not note that children suffering serious symptoms from COVID-19 is incredibly rare. Children account for only 13% of COVID-19 CASES, per the Mayo Clinic, and of those who do contract the China-originated virus, “Many of them have no symptoms,” reports Harvard Health. “Those that do get sick tend to experience milder symptoms such as low-grade fever, fatigue, and cough.”…


Cases are not infections, they are PCR signals.   Infections require manifestly  uncontrolled viral reproduction in the human body i.e. illness.   By that standard, the rate of covid infections among children must be minuscule.   Unfortunately, the rate of adult-administered child abuse is off the charts and hugely destructive, especially when it has government support.
